Brain organoids, derived from pluripotent stem cells, mimic key aspects of the human brain and are valuable for studying neurodevelopment and disease. However, current methods primarily generate neural components and lack a crucial vascular system, which is essential for brain functions. Vascularization of brain organoids is important for studying diseases related to cerebral microcirculation. Efforts to create vascularized brain organoids include fusion of vascular and brain organoids, in vivo organoid transplantation, and microfluidic devices. This review summarizes these approaches and emphasizes the significance of studying the neurovascular unit for understanding and treating neurological disorders.
